Jamu pegal linu, atau dikenal sebagai jamu cabe puyang, merupakan obat tradisional yang digunakan untuk meredakan nyeri otot dan melancarkan peredaran darah. Salah satu bahan utamanya adalah cabe jawa (Piper retrofractum) yang mengandung senyawa aktif piperin. Penelitian ini bertujuan untuk memvalidasi dalam dan menetapan kadar piperin pada serbuk jamu pegal linu metode High Performance Liquid Chromatography (HPLC). Parameter validasi yang diuji meliputi akurasi, presisi, linearitas, batas deteksi (LOD), dan batas kuantifikasi (LOQ). Validasi metode dan penetapan kadar piperin dilakukan dengan metode HPLC fase terbalik dengan kolom C18, fase gerak metanol:air (9:1), laju alir 1,0 ml/menit, serta deteksi dengan panjang gelombang 341 nm. Hasil validasi menunjukkan bahwa metode ini memiliki akurasi sebesar 99,03%, presisi (%RSD) sebesar 1,36%, dan linearitas sangat baik dengan nilai korelasi (r) sebesar 0,999 pada rentang konsentrasi 1; 2,5; 5; 7,5; dan 10 μg/mL. Nilai LOD dan LOQ masing-masing diperoleh sebesar 0,117 µg/mL dan 0,390 µg/mL. Kadar piperin dalam sampel serbuk jamu pegal linu yang dianalisis adalah sebesar sampel A sebesar 1,420 ± 0,054 mg/gram, sampel B sebesar 0,077 ± 0,011 mg/gram, dan sampel C sebesar 0,266±0,008 mg/gram. Hasil penelitian menunjukkan bahwa metode HPLC telah memenuhi parameter yang dipersyaratkan dan dinyatakan valid untuk analisis piperin dalam sediaan jamu.
